pSEVA341 is compiled with a chloramphenicol antibiotic resistance, pRO1600/ColE1 origin from pUCP24 with a high copy number (~25-40), and an MSC. This backbone is golden-gate compatible with BsaI. The promoter region is designed to have a high transcription rates and the ribosome binding site is designed to have a high translation rate to produce high levels of protein from the inserted gene. The RBS is consistent with the RBS found in the riboJ-RBS part (BBa_K4215001), which allows for controlled bicistronic gene expression.
